PF-05089771 + pregabalin is a combination of two investigational and approved drugs studied for the treatment of neuropathic pain, particularly painful diabetic peripheral neuropathy. PF-05089771 is a potent and selective blocker of the sodium channel protein type 9 subunit alpha (Nav1.7), which is predominantly expressed in nociceptive and sympathetic neurons. Pregabalin is an anticonvulsant that binds to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system, modulating neurotransmitter release to reduce pain signaling. The combination was evaluated in clinical trials to assess potential additive or synergistic analgesic effects; however, studies did not demonstrate significant additional benefit over monotherapy[2][3][7].
