Overview

PF-05175157 is an orally available, small molecule inhibitor of acetyl-CoA carboxylase (ACC), targeting both ACC1 and ACC2 isoforms. It was developed by Pfizer as a potential treatment for type 2 diabetes mellitus and has also been studied in the context of acne vulgaris. The drug acts by inhibiting ACC, a key enzyme involved in fatty acid synthesis and metabolism, thereby modulating lipid homeostasis and glucose metabolism. In preclinical studies, it demonstrated broad-spectrum antiviral activity against viruses such as West Nile virus, dengue virus, and Zika virus through host-targeted mechanisms. Clinical development reached phase II before being discontinued due to safety concerns related to increased risk of myocardial injury following ischemic events[1][3][4][5].
