PF-05212389 (also known as OAP-189) is a PEGylated analog of the peptide hormone oxyntomodulin, developed by Pfizer for the treatment of obesity and type 2 diabetes mellitus. It functions as a dual agonist, targeting both the glucagon-like peptide-1 (GLP-1) receptor and the glucagon receptor. By activating the GLP-1 receptor, the drug aims to enhance insulin secretion and suppress appetite, while its action on the glucagon receptor is intended to increase energy expenditure and promote weight loss. Clinical development included Phase 1 studies evaluating its safety, gastrointestinal tolerability (specifically nausea and vomiting), and pharmacokinetics, particularly in combination with metformin. However, development of this specific candidate was discontinued by Pfizer in favor of other metabolic disease assets.
