Drug intelligence / Profile preview
PF-05231023 (also known as CVX-343) is a long-acting, antibody-conjugated fibroblast growth factor 21 (FGF21) mimetic developed by Pfizer and CovX. It consists of two engineered human FGF21 molecules covalently linked to the Fab region of a humanized IgG1 scaffold antibody (CVX-2000). PF-05231023 acts as an agonist of the FGF21 receptor complex, which comprises the co-receptor beta-klotho (KLB) and fibroblast growth factor receptors (primarily FGFR1c, but also FGFR2c and FGFR3c). It was developed for the treatment of type 2 diabetes mellitus, obesity, and hyperlipidemia. Although it demonstrated favorable lipid-lowering effects and modest weight loss in clinical trials, its development was discontinued due to safety concerns, including increases in blood pressure and heart rate, as well as shifts in bone turnover biomarkers.
