PF-05251749 (BIIB118) is an orally administered small molecule inhibitor of casein kinase 1 delta (CK1δ) and casein kinase 1 epsilon (CK1ε), originally developed by Pfizer and later acquired by Biogen. The drug was investigated for the treatment of Alzheimer's disease, Parkinson's disease, circadian rhythm sleep disorders, and behavioral symptoms such as Sundowning in Alzheimer's disease. CK1δ/ε are key regulators of the circadian clock; their inhibition is hypothesized to correct circadian rhythm disruptions that contribute to neurodegenerative symptoms. Clinical development reached Phase 1 trials for these indications but has since been discontinued[1][2][7].
