Overview
PF-06263507 is an investigational antibody-drug conjugate (ADC) developed for the treatment of advanced solid tumors. It consists of a humanized IgG1 monoclonal antibody targeting the 5T4 oncofetal antigen (also known as trophoblast glycoprotein or TPBG), which is overexpressed in various cancers. The antibody is covalently linked via a non-cleavable maleimidocaproyl (mc) linker to monomethylauristatin F (MMAF), a potent microtubule-disrupting cytotoxic agent. Upon binding to 5T4-expressing tumor cells, the ADC is internalized and MMAF disrupts microtubule dynamics leading to cell death. Preclinical studies demonstrated specific binding and potent antitumor activity in xenograft models. Clinical trials evaluated its safety and pharmacokinetics in patients with advanced solid malignancies[3][6][5].
