Overview

PF-06266047 is a small molecule phosphodiesterase 4 (PDE4) inhibitor developed by Pfizer. It selectively inhibits PDE4 isoforms and is brain penetrant with sparing of the PDE4D isozyme to potentially reduce dose-limiting side effects such as nausea and vomiting commonly associated with PDE4 inhibitors. The drug has been investigated primarily for the treatment of schizophrenia and shows anti-inflammatory effects in preclinical models. It has demonstrated efficacy in improving motor function and reducing neuroinflammation in rodent models of juvenile Batten disease by restoring cAMP levels in the brain. Clinical development has included Phase I trials assessing safety, tolerability, and pharmacokinetics in healthy volunteers[1][2][3][6].
