Overview

PF-06291874 is a highly potent, non-peptide, orally active small molecule glucagon receptor antagonist developed by Pfizer. It is being investigated for the treatment of type 2 diabetes mellitus (T2DM). The drug works by blocking the action of glucagon at its receptor (glucagon receptor/GCGR), thereby reducing hepatic glucose production and lowering blood glucose levels. Clinical studies have shown that PF-06291874 significantly lowers HbA1c and fasting plasma glucose in patients with T2DM on background metformin therapy, with a low risk of hypoglycemia. The most common side effects observed were small increases in liver enzymes and minor changes in blood pressure and lipids[1][3][6][7].
