Overview

PF-06293620 is a humanized IgG2Δa monoclonal antibody developed by Pfizer as a glucagon receptor antagonist for the treatment of type 2 diabetes mellitus. It binds to and blocks the human glucagon receptor, thereby inhibiting glucagon signaling. This mechanism reduces hyperglycemia by normalizing dysregulated hepatic glucose production, independent of pancreatic beta cell function. The drug was administered subcutaneously in clinical trials and demonstrated robust lowering of fasting plasma glucose (FPG) and HbA1c in subjects with type 2 diabetes mellitus on stable metformin therapy. Development reached Phase 1 clinical trials but has not progressed further[1][3][4].
