Overview

PF-06647263 is a novel antibody-drug conjugate (ADC) developed by Pfizer for the treatment of advanced solid tumors, particularly triple-negative breast cancer (TNBC) and ovarian cancer. The drug consists of a humanized monoclonal antibody targeting ephrin-A4 (EFNA4), which is overexpressed in several tumor types, conjugated via a hydrazone cleavable linker to calicheamicin, a potent DNA-damaging cytotoxic agent. The mechanism of action involves targeted delivery of calicheamicin to EFNA4-expressing tumor cells, resulting in DNA damage and cell death. Preclinical studies demonstrated significant antitumor activity in xenograft models. In clinical trials, the drug was administered intravenously and showed manageable safety with preliminary evidence of limited antitumor activity; however, development was discontinued after phase I due to limited efficacy[1][2][3][5].
