PF-06730512 is a recombinant fusion protein consisting of the first two immunoglobulin domains of human Roundabout Guidance Receptor 2 (ROBO2) fused to a human immunoglobulin G1 crystallized fragment (Fc). It was developed by Pfizer for the treatment of focal segmental glomerulosclerosis (FSGS), a kidney disorder characterized by proteinuria. The drug acts as a neutralizing ligand trap that binds to slit guidance ligand 2 (SLIT2), preventing its interaction with native ROBO2 on podocytes in the glomeruli. By inhibiting SLIT2/ROBO2 signaling, PF-06730512 is designed to stabilize podocyte structure, enhance actin polymerization and adhesion, and reduce protein leakage into urine. Clinical development reached Phase 2 trials but has since been discontinued for FSGS[1][5][6].
