Drug intelligence / Profile preview
PF-06730512 (ROBO2-IgG-Fc fusion protein) is a recombinant fusion protein consisting of the first two immunoglobulin domains of the human roundabout guidance receptor 2 (ROBO2) fused to a human immunoglobulin G1 (IgG1) Fc region. Developed by Pfizer in collaboration with Boston University and Boston Medical Center, PF-06730512 acts as a ligand trap (antagonist) that binds to slit guidance ligand 2 (SLIT2), preventing its interaction with endogenous ROBO2 on podocytes. This blockade is designed to stabilize podocyte adhesion and actin polymerization, thereby maintaining the structural integrity of the glomerular filtration barrier. PF-06730512 was evaluated in Phase 2 clinical trials for the treatment of focal segmental glomerulosclerosis (FSGS) and nephrotic syndrome, but development was discontinued in late 2023 due to a lack of clinical efficacy.
