Overview

PF-06730512 is a recombinant ROBO2-Fc fusion protein designed to act as a SLIT ligand trap. It consists of the first two immunoglobulin domains of the roundabout guidance receptor 2 (ROBO2) fused to a mutated human immunoglobulin G1 (IgG1) Fc region. By binding to endogenous slit guidance ligand 2 (SLIT2), PF-06730512 prevents the interaction of SLIT2 with native ROBO2 on the basal surface of podocytes. This blockade inhibits the downstream signaling that normally destabilizes the slit diaphragm and reduces podocyte adhesion, thereby promoting podocyte actin polymerization and structural integrity. Developed by Pfizer, PF-06730512 was evaluated in a Phase II clinical trial (PODO) for focal segmental glomerulosclerosis (FSGS) but development was discontinued due to a lack of efficacy.
