Overview

PF-06747143 is a humanized IgG1 monoclonal antibody antagonist of CXCR4, developed to disrupt the CXCR4/CXCL12 signaling axis, which is involved in the retention of malignant cells within the protective bone marrow niche and contributes to therapy resistance. PF-06747143 mobilizes tumor cells into the circulation, sensitizing them to chemotherapeutic agents, and induces tumor cell death via Fc-mediated effector function. Cytarabine and daunorubicin are standard-of-care cytotoxic chemotherapies for AML, targeting DNA synthesis and topoisomerase II, respectively[1][2][3]. The rationale for their combination is to synergistically enhance antitumor efficacy by mobilizing and eliminating resistant leukemic cells[1].
