Overview

PF-06835375 is a humanized, afucosyl immunoglobulin G1 (IgG1) monoclonal antibody developed by Pfizer. It acts as a selective antagonist of the C-X-C chemokine receptor type 5 (CXCR5), which is expressed on B cells, T follicular helper (Tfh) cells, and circulating Tfh-like (cTfh) cells. The drug depletes CXCR5-positive B and Tfh cells and antagonizes C-X-C motif chemokine ligand 13-dependent signaling, thereby modulating immune responses implicated in autoimmune diseases. Its primary indications under investigation include idiopathic thrombocytopenic purpura (ITP), systemic lupus erythematosus (SLE), and rheumatoid arthritis[1][4][7]. As of early 2024, it is in phase II clinical trials for ITP and phase I trials for SLE and rheumatoid arthritis[1].
