Drug intelligence / Profile preview
PF-06869206 is an orally bioavailable, highly selective small molecule inhibitor of the renal sodium-phosphate cotransporter 2a (NaPi-IIa, encoded by the *SLC34A1* gene). Developed by Pfizer, the compound selectively blocks phosphate reabsorption in the proximal renal tubules, promoting urinary phosphate excretion (phosphaturia) and subsequently lowering circulating phosphate and parathyroid hormone (PTH) levels. It exhibits high selectivity for NaPi-IIa over other phosphate transporters such as NaPi-IIb, NaPi-IIc, PiT-1, and PiT-2. PF-06869206 has been widely utilized as a chemical tool in preclinical research to investigate phosphate homeostasis, chronic kidney disease-mineral and bone disorder (CKD-MBD), and acute kidney injury (AKI).
