Overview

PF-07062119 is a T cell-engaging bispecific antibody developed by Pfizer for the treatment of advanced gastrointestinal cancers, including colorectal, gastric, and esophageal adenocarcinomas. The drug is designed to redirect immune cells (T cells) to recognize and kill cancer cells by simultaneously binding to CD3 on T cells and Guanylyl Cyclase C (GUCY2C) on tumor cells. GUCY2C is widely expressed in gastrointestinal malignancies but has limited expression in normal tissues, allowing for selective targeting of tumors. Preclinical studies demonstrated potent tumor-selective efficacy and manageable toxicity. Clinical evaluation has focused on safety, tolerability, pharmacokinetics, pharmacodynamics, and preliminary anti-tumor activity as both a single agent and in combination with anti-PD1 (sasanlimab/PF-06801591) or anti-VEGF (bevacizumab-Pfizer). Administration is via subcutaneous injection[2][3][4][5].
