PF-07062119 is an antibody-drug conjugate (ADC) developed by Pfizer, comprising a humanized IgG1 monoclonal antibody directed against programmed cell death 1 ligand 1 (PD-L1) conjugated to the cytotoxic microtubule-disrupting agent monomethyl auristatin E (MMAE). The conjugation is achieved via a protease-cleavable valine-citrulline (vc) linker. PF-07062119 is designed to selectively bind to PD-L1-expressing tumor cells, leading to internalization of the ADC and subsequent lysosomal cleavage of the linker. This release of MMAE into the cytoplasm inhibits tubulin polymerization, resulting in cell cycle arrest and apoptosis. Beyond direct cytotoxicity, the targeting of PD-L1 may also interfere with the PD-1/PD-L1 signaling axis, potentially enhancing anti-tumor immune responses. Clinical development was focused on advanced solid tumors, including non-small cell lung cancer (NSCLC), but the program was terminated by the sponsor for strategic reasons.
