Overview

PF-07224826 is an orally bioavailable small molecule inhibitor targeting cyclin-dependent kinases 2, 4, and 6 (CDK2/4/6). It was developed by Pfizer for the potential treatment of advanced solid tumors, including breast cancer (notably hormone receptor-positive/HER2-negative metastatic breast cancer), ovarian cancer (platinum-resistant), triple negative breast cancer (TNBC), liposarcoma, endometrial cancer, non-small cell lung cancer (NSCLC), and other advanced malignancies. The drug acts by inhibiting CDK2/4/6 activity to disrupt cell cycle progression in tumor cells. Clinical development included evaluation as a single agent and in combination with endocrine therapy such as fulvestrant. The primary route of administration is oral dosing on a continuous daily schedule[1][2][3][4][5].
