PF-07225570 + sasanlimab

PF-07225570 + sasanlimab is an investigational combination therapy being developed for the treatment of non-muscle invasive bladder cancer (NMIBC), specifically in the high-risk, BCG-naïve population. PF-07225570 is a novel immunostimulatory agent whose precise mechanism of action has not been fully disclosed but is known to act by stimulating certain immune cells. Sasanlimab is a subcutaneously administered monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, functioning as a checkpoint inhibitor to reactivate T-cell mediated anti-tumor immunity. The combination is under investigation by Pfizer, which has conducted both phase 1 and phase 3 clinical trials. In phase 1, PF-07225570 was studied alone and in combination with sasanlimab in recurrent NMIBC. In phase 3 (CREST trial), sasanlimab in combination with BCG demonstrated a significant improvement in event-free survival in BCG-naïve, high-risk NMIBC compared to standard of care.
