Overview

PF-07258669 is an orally available small molecule drug developed by Pfizer. It acts as a selective antagonist of the melanocortin-4 receptor (MC4R), a G protein-coupled receptor involved in regulating energy homeostasis and somatic growth. By blocking MC4R, PF-07258669 is intended to increase appetite and promote weight gain, making it a potential treatment for unintended weight loss and malnutrition, particularly in older adults. The drug is currently in Phase 1 clinical trials for malnutrition and has also been investigated for anorexia. As of March 2025, it remains an investigational new molecular entity with no reported orphan drug status.
