Overview

PF-07284892 (also known as ARRY-558) is an investigational, orally active small molecule inhibitor of SHP2 (Src homology region 2 domain-containing phosphatase 2, also known as PTPN11). It acts as an allosteric inhibitor by binding to and inhibiting the activity of SHP2, thereby blocking downstream signaling through receptor tyrosine kinases (RTKs) and the RAS/MAPK pathway. This mechanism is designed to prevent or overcome resistance to targeted therapies in oncogene-driven solid tumors by disrupting bypass signaling pathways that drive tumor growth. Preclinical and early clinical studies have shown that PF-07284892 can sensitize tumors with acquired resistance to other targeted agents when used in combination therapy. The drug was originally discovered by Array BioPharma and is being developed by Pfizer for use in oncology, particularly for patients with advanced solid tumors harboring oncogenic drivers who have progressed on prior targeted therapies.
