Overview

PF-07328948 is an orally administered, small molecule inhibitor of branched-chain ketoacid dehydrogenase kinase (BCKDK or BDK), developed by Pfizer for the treatment of heart failure, particularly heart failure with preserved ejection fraction. By inhibiting BCKDK, PF-07328948 enhances the catabolism of branched-chain amino acids (BCAAs) through degradation of the BCKDH enzyme complex. This mechanism improves metabolic and cardiac endpoints in preclinical rodent models and lowers plasma biomarkers such as BCAAs and branched-chain ketoacids in vivo. The drug has demonstrated good tolerability and a pharmacokinetic profile suitable for once-daily dosing in Phase 1 clinical trials[1][3][4][5][6][8].
