Overview

PF-07329640 is a tetravalent monoclonal antibody developed by Pfizer as an agonist of the lymphotoxin beta receptor (LTβR, also known as TNFRSF3). The drug was designed to activate LTβR signaling, with the aim of inducing tertiary lymphoid structure formation and maturation in solid tumors—a novel immunotherapeutic approach. It was investigated primarily for advanced or metastatic solid tumors, including melanoma, non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and urothelial carcinoma. The agent was tested both as monotherapy and in combination with bevacizumab or sasanlimab. Clinical development reached phase 1/2 trials but was terminated after limited enrollment due to strategic business decisions[2][5][6].
