PF-07852352 is a self-amplifying ribonucleic acid (saRNA) vaccine candidate developed by Pfizer for the prevention of influenza. Unlike conventional mRNA vaccines, saRNA vaccines incorporate a viral replicase enzyme complex (typically derived from an alphavirus backbone) that allows the RNA to replicate itself once inside the host cell cytoplasm. This self-amplification mechanism is designed to drive higher and more sustained expression of the encoded influenza antigens, potentially enabling lower dose requirements to achieve protective immunity. PF-07852352 was evaluated in Phase 1 clinical trials (such as NCT05052697) alongside multiple other saRNA constructs to assess safety, tolerability, and immunogenicity in healthy adults compared to standard quadrivalent influenza vaccines.
