Overview

PF-07921585 is a novel investigational biologic drug developed by Pfizer for the treatment of advanced solid tumors, including non-small cell lung cancer, metastatic colorectal cancer, renal cell carcinoma, bladder cancer, melanoma, urothelial carcinoma, and head and neck squamous cell carcinoma. It is an interleukin 12 (IL-12) mutein fusion protein designed as a partial agonist of IL-12 with additional PD-1 inhibitory activity. The drug acts by stimulating the interleukin 12 receptor (IL-12R) to enhance anti-tumor immune responses while also inhibiting programmed cell death protein 1 (PD-1), potentially overcoming tumor-induced immunosuppression. PF-07921585 is administered intravenously or subcutaneously every three weeks and may be used alone or in combination with other anti-cancer agents such as sasanlimab (an anti-PD1 antibody). As of early 2025, it remains in Phase I clinical trials for multiple advanced/metastatic solid tumor indications[3][4][5][6][7].
