Drug intelligence / Profile preview
**PF-07976016 + liraglutide** is an investigational combination regimen consisting of PF-07976016, an orally bioavailable small-molecule antagonist of the gastric inhibitory polypeptide receptor (GIPR), and liraglutide, a subcutaneous peptide agonist of the glucagon-like peptide-1 receptor (GLP-1R). Developed by Pfizer, PF-07976016 was designed as a first-in-class selective small-molecule GIPR antagonist to evaluate whether blocking GIPR signaling could provide additive weight loss when combined with GLP-1R agonism. The combination was evaluated in a Phase 2a clinical trial (NCT06717425) for the treatment of adults with obesity. However, in August 2026, Pfizer announced the discontinuation of PF-07976016's development following a review of the Phase 2a data on the liraglutide background.
