Drug intelligence / Profile preview
PF-07976016 + PF-08642534 is an investigational oral combination therapy that was being developed by Pfizer for the treatment of obesity. The combination consists of PF-07976016, an orally bioavailable small molecule antagonist of the gastric inhibitory polypeptide receptor (GIPR), and PF-08642534 (also known as YP05002), an oral small molecule glucagon-like peptide-1 (GLP-1) receptor agonist licensed from YaoPharma. By combining GIPR antagonism with GLP-1R agonism, the therapy aimed to achieve enhanced weight loss compared to GLP-1R agonism alone. However, in August 2026, Pfizer announced the discontinuation of PF-07976016 following a review of Phase 2a clinical data, which effectively halted the development of this specific combination.
