Overview
PF-08046050 is an investigational antibody-drug conjugate (ADC) targeting CEA cell adhesion molecule 5 (CEACAM5), a glycoprotein highly expressed in several epithelial tumors such as colorectal cancer, pancreatic ductal adenocarcinoma, gastric carcinoma, gastroesophageal junction adenocarcinoma, and non-small cell lung cancer. The ADC consists of a humanized IgG1 monoclonal antibody directed against CEACAM5 and is conjugated to a novel topoisomerase I inhibitor payload (aminomethyl-10,11-methylenedioxycamptothecin; AMDCPT). This design enables preferential release of the cytotoxic payload within target tumor cells expressing CEACAM5. The drug is being developed by Pfizer following its acquisition of Seagen and in partnership with Sanofi for the monoclonal antibody component[1][4][9][10]. It is currently under investigation in Phase I clinical trials for advanced solid tumors.
