PF-08052666 (also known as SGN-MesoC2 or HBM9033) is an antibody-drug conjugate (ADC) being developed by Pfizer (following its acquisition of Seagen) for the treatment of advanced solid tumors. The drug targets mesothelin, a cell-surface glycoprotein that is highly expressed in various cancers, including mesothelioma, ovarian cancer, and pancreatic cancer, while having limited expression in normal tissues. PF-08052666 consists of a humanized monoclonal antibody against mesothelin conjugated to a cytotoxic topoisomerase I inhibitor payload via a protease-cleavable linker. It is currently being evaluated in Phase 1 clinical trials to assess its safety, pharmacokinetics, and preliminary antitumor activity in patients with unresectable or metastatic solid tumors that have progressed on standard therapies.
