PF-08103402 is an investigational, orally bioavailable small molecule antagonist of the G protein-coupled receptor 44 (GPR44), also known as the chemoattractant receptor-homologous molecule expressed on Th2 cells (CRTh2). Developed by Pfizer, it is currently in Phase 1 clinical development for the treatment of asthma. GPR44 is a high-affinity receptor for prostaglandin D2 (PGD2), a primary mediator released by mast cells during allergic inflammation. Activation of GPR44 triggers the recruitment and activation of Th2 cells, eosinophils, and basophils, which are central to the pathophysiology of allergic airway diseases. By selectively antagonizing GPR44, PF-08103402 aims to inhibit these inflammatory pathways and provide a novel therapeutic option for patients with mild-to-moderate asthma.
