Overview

PF-08154225 is an investigational, non-viral, mRNA-based chimeric antigen receptor (CAR) T-cell therapy being developed by Pfizer for the treatment of severe B-cell mediated autoimmune diseases. The therapy involves engineering T cells to express a CAR that targets the CD19 antigen, a protein widely expressed on the surface of B lymphocytes, including those responsible for producing pathogenic autoantibodies. By utilizing mRNA technology for transient CAR expression rather than permanent viral-vector integration, PF-08154225 may offer a more controlled B-cell depletion profile and the potential for multiple dosing. The primary goal of the therapy is to achieve a deep depletion of the B-cell compartment to reset the immune system in patients with systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), idiopathic inflammatory myositis (IIM), and systemic sclerosis (SSc). It is currently being evaluated in a Phase 1/2 clinical trial (NCT06344104) to assess its safety, tolerability, and preliminary efficacy.
