Overview

PF-08653944 (formerly MET-097i) is an ultra-long-acting, fully biased glucagon-like peptide-1 (GLP-1) receptor agonist being developed by Pfizer for the treatment of obesity and overweight. Acquired through the $10 billion purchase of Metsera in late 2025, the drug is designed for subcutaneous injection with potential for both weekly and monthly dosing. In the Phase 2b VESPER-3 study, PF-08653944 demonstrated a placebo-adjusted mean weight loss of 12.3% at 28 weeks. Pfizer is initiating a broad Phase 3 program consisting of 10 trials to evaluate the drug as a monotherapy and in combination with other metabolic agents, such as amylin analogs (PF-08653945) and GIP receptor agonists (PF-08654696), aiming for regulatory approvals starting in 2028.
