Drug intelligence / Profile preview
PF-08654696 + berobenatide (PF-08653944) is an investigational combination therapy that was developed for chronic weight management and obesity. The combination consists of PF-08654696, an ultra-long-acting glucose-dependent insulinotropic polypeptide receptor (GIPR) agonist (previously MET-034i), and berobenatide (PF-08653944), an ultra-long-acting, fully biased glucagon-like peptide-1 receptor (GLP-1R) agonist (previously MET-097i). The assets originated from research at Imperial College London and its spinout Zihipp, were subsequently developed by Metsera, and were acquired by Pfizer in late 2025. Berobenatide is designed to selectively activate cyclic adenosine monophosphate (cAMP) signaling over beta-arrestin recruitment, which prevents receptor internalization and extends its half-life to support once-monthly subcutaneous dosing. In November 2026, Pfizer announced the discontinuation of the Phase 1 combination program, terminating further development of the GIPR agonist PF-08654696, while continuing to advance berobenatide as a monotherapy in Phase 3 trials.
