It functions as an agonist of the GDNF family receptor alpha-like (GFRAL), which is the high-affinity receptor for Growth Differentiation Factor 15 (GDF15). By activating the GFRAL-RET signaling complex in the hindbrain, specifically within the area postrema and nucleus of the solitary tract, the drug is intended to reduce food intake and promote weight loss. PF-08656795 is engineered as a long-acting therapy to allow for infrequent dosing. It is currently in Phase 1 clinical development to evaluate its safety, tolerability, and pharmacokinetics in individuals with obesity.
