Overview

PF-3512676 (also known as CPG 7909) is a synthetic oligodeoxynucleotide that acts as an agonist of Toll-like receptor 9 (TLR9), stimulating the immune system and enhancing immune activation in multiple tumor types. Erlotinib is a small molecule tyrosine kinase inhibitor that targets the epidermal growth factor receptor (EGFR), blocking its activity and thereby inhibiting cancer cell proliferation and survival. The combination of PF-3512676 with erlotinib was investigated for potential synergistic effects in treating advanced non-small cell lung cancer (NSCLC), particularly in EGFR-positive patients, based on their complementary mechanisms—immune activation by PF-3512676 and EGFR inhibition by erlotinib. However, clinical trials did not demonstrate improved progression-free survival with the combination compared to erlotinib alone[2].
