Overview

This is a combination regimen of four agents: PF-3512676 (also known as CpG 7909), paclitaxel, carboplatin, and bevacizumab. PF-3512676 is a synthetic oligodeoxynucleotide that acts as an agonist of Toll-like receptor 9 (TLR9), activating plasmacytoid dendritic cells and B lymphocytes to stimulate innate and adaptive immune responses against tumors. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division, carboplatin is a platinum-based chemotherapy that causes DNA crosslinking and apoptosis, and bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), blocking tumor angiogenesis. This combination was investigated for advanced non-small cell lung cancer (NSCLC), but addition of PF-3512676 to standard cytotoxic chemotherapy regimens (including paclitaxel and carboplatin, with or without bevacizumab) failed to show added clinical benefit and resulted in increased toxicity, especially sepsis and myelosuppression. Pfizer discontinued further development of the combination for lung cancer following interim analyses in phase III trials.
