Overview

PF-5177624 (also known as PF-05177624) is a potent and selective small molecule inhibitor of 3-phosphoinositide-dependent kinase-1 (PDK1, also known as PDPK1). Developed by Pfizer, it targets the PI3K/AKT signaling pathway, which is frequently dysregulated in various cancers, including breast cancer. By inhibiting PDK1, PF-5177624 blocks the phosphorylation of downstream substrates such as AKT (at Thr308) and p70S6K, thereby inhibiting cell proliferation, cell cycle progression, and cell transformation, and inducing apoptosis in cancer cells. Although investigated preclinically for the treatment of solid tumors and breast cancer, development of the compound appears to have been discontinued.
