Overview

PF4950834 is a small molecule inhibitor of Rho-associated protein kinase (ROCK) developed by Pfizer. It has been investigated for its potential therapeutic role in chronic obstructive pulmonary disease (COPD). In preclinical studies using macrophages from COPD patients, PF4950834 was shown to enhance the defective efferocytosis (clearance of apoptotic cells) without impairing the phagocytosis or intracellular killing of bacteria such as *Streptococcus pneumoniae* and *Haemophilus influenzae*. By inhibiting the ROCK pathway, the compound reduces the phosphorylation of myosin light chain, which is implicated in the regulation of the macrophage cytoskeleton and inflammatory responses.
