Overview

PfAMA1-FVO is a recombinant protein vaccine candidate designed to prevent *Plasmodium falciparum* malaria. It consists of the ectodomain (amino acids 25–545) of the Apical Membrane Antigen 1 (AMA1) derived from the FVO (Falcetti/Vietnam/Oak Knoll) strain of the parasite. Expressed in the yeast *Pichia pastoris*, the protein is typically formulated with an adjuvant such as Alhydrogel. AMA1 is a microneme protein essential for the invasion of host erythrocytes; it interacts with Rhoptry Neck Protein 2 (RON2) to facilitate the formation of a moving junction during parasite entry. The vaccine aims to induce high titers of neutralizing antibodies that block this interaction, thereby inhibiting the parasite's life cycle at the blood stage.
