PFL-13 is a small molecule antiviral lead compound identified through a high-throughput screen (HTS) of over 40,000 compounds for the prevention of Human Papillomavirus (HPV) infections. Developed by researchers at the University of Wisconsin-Madison, PFL-13 was sourced from the Chembridge DIVERSet library (Chembridge No. 6228481). It exhibits potent, submicromolar inhibitory activity (IC50 < 0.1 µM) against multiple HPV genotypes, including high-risk HPV16 and HPV31, as well as low-risk HPV11. Mechanistic studies indicate that PFL-13 acts at an early post-entry stage of the viral life cycle, specifically within the first 12 hours of infection, without preventing the initial internalization of the virus. Due to its high potency and relatively low cytotoxicity (CD50 ~30 µM), it is considered a promising candidate for development as a topical microbicide to prevent sexually transmitted HPV infections.
