Overview

PFL-14 is a small molecule antiviral lead compound identified through high-throughput screening for the prevention of Human Papillomavirus (HPV) infection. It demonstrates potent, broad-spectrum activity against multiple HPV genotypes, including high-risk (HPV16, HPV31) and low-risk (HPV11) types, with submicromolar IC50 values and minimal cytotoxicity. Mechanistically, PFL-14 acts at an early stage of the viral life cycle, specifically at a post-entry step within the first 12 hours of infection, without preventing the initial internalization of the virus into the host cell. It was discovered by researchers at the University of Wisconsin-Madison and is envisioned as a potential topical microbicide to reduce the incidence of HPV-associated cancers.
