Overview

**PfRipr5** is the most immunogenic fragment (C720-D934) of the *Plasmodium falciparum* merozoite Rh5 interacting protein (PfRipr), a highly conserved component of the PfRipr/CyRPA/Rh5 complex essential for erythrocyte invasion during the asexual blood stage of malaria. Antibodies against PfRipr5 potently inhibit merozoite invasion by blocking PfRipr interactions with PfRh5 and the erythrocyte receptor SEMA7A. Developed as a recombinant protein vaccine candidate produced in insect cells (High Five), it demonstrates strong immunogenicity when adjuvanted (e.g., with CAF®01 or SA-1), eliciting growth-inhibitory antibodies in preclinical models, with ongoing efforts to establish cGMP manufacturing and advance to phase I/IIa trials.[2][3][5][7][11]
