Overview

Pfs230D1-CRM197 is a recombinant protein-based malaria transmission-blocking vaccine candidate. It consists of the domain 1 fragment of the *Plasmodium falciparum* sexual stage surface antigen Pfs230 (Pfs230D1) chemically conjugated to CRM197, a non-toxic mutant of diphtheria toxin used as a carrier protein. The vaccine is designed to elicit antibodies that target gametes of the malaria parasite in humans, thereby blocking parasite development in mosquitoes and interrupting transmission. The mechanism involves induction of antibodies against Pfs230, which are thought to fix complement and lyse gametes within the mosquito midgut after ingestion from an infected human host[6][8]. This approach aims to reduce or eliminate malaria transmission within communities. Clinical studies have shown that conjugation with CRM197 enhances immunogenicity compared to other carriers[4]. The vaccine has been evaluated in combination with R21/Matrix-M for broader protection against clinical malaria and reduction of community-level transmission[3].
