Drug intelligence / Profile preview
Pfs230D1-E2p is a preclinical single-component nanoparticle vaccine candidate designed to block the transmission of *Plasmodium falciparum* malaria. Developed by the National Institute of Allergy and Infectious Diseases (NIAID), the vaccine is a genetic fusion of Domain 1 of the *P. falciparum* gametocyte surface protein Pfs230 (Pfs230D1) to the catalytic domain of dihydrolipoyl acetyltransferase (E2p) from *Geobacillus stearothermophilus*. This fusion self-assembles into a stable, homogenous 60-copy nanoparticle that repetitively displays the Pfs230D1 antigen. By presenting a potent human transmission-blocking conformational epitope, Pfs230D1-E2p elicits a robust and durable antibody response that prevents the parasite from infecting the mosquito vector, thereby breaking the malaria transmission cycle.
