Drug intelligence / Profile preview
Pfs230D1-EPA + AS01 is an investigational malaria transmission-blocking vaccine (TBV) candidate developed by the National Institute of Allergy and Infectious Diseases (NIAID). It consists of the recombinant first domain of the Plasmodium falciparum gamete surface protein Pfs230 (Pfs230D1) chemically conjugated to a detoxified Pseudomonas aeruginosa ExoProtein A (EPA) carrier protein, formulated with GlaxoSmithKline's (GSK) liposome-based adjuvant system AS01. The vaccine is designed to elicit antibodies against Pfs230, which are ingested by mosquitoes during a blood meal. In the presence of human complement, these antibodies target and lyse the extracellular gametes of P. falciparum in the mosquito midgut, thereby blocking parasite development and preventing transmission to other humans. It has been evaluated in Phase 1 and Phase 2 clinical trials in malaria-endemic regions such as Mali.
