Drug intelligence / Profile preview
Pfs25-IMX313 + Matrix-M is an investigational malaria transmission-blocking vaccine candidate. It consists of the recombinant protein antigen Pfs25 fused to IMX313, a heptamerization domain derived from chicken complement inhibitor C4-binding protein, which enhances immunogenicity by forming nanoparticle-like structures. The vaccine is formulated with the saponin-based adjuvant Matrix-M to further boost immune responses. The primary mechanism is to induce antibodies against the sexual-stage surface protein Pfs25 of *Plasmodium falciparum*, which are taken up by mosquitoes during a blood meal and block parasite development in the mosquito midgut, thereby preventing onward transmission of malaria. This approach does not directly protect vaccinated individuals from clinical malaria but aims to reduce community-level transmission rates[1][5][6]. The vaccine has been developed and tested primarily by researchers at the University of Oxford and associated partners, with clinical trials conducted in Africa (e.g., Tanzania, Burkina Faso) and preclinical studies supporting its safety and immunogenicity[2][7][8].
