Pfs25-Pfs25 Conjugate is an investigational malaria transmission-blocking vaccine candidate composed of multiple molecules of the Plasmodium falciparum surface antigen Pfs25 chemically linked to itself, forming a polymeric conjugate. The rationale for this design is to enhance immunogenicity by increasing molecular size and epitope density, thereby improving antibody responses compared to monomeric or unconjugated forms. Preclinical studies in mice demonstrated that these conjugates, typically consisting of 2–10 Pfs25 molecules (50–250 kDa), induce robust anti-Pfs25 antibody titers and significant transmission-blocking activity as measured by reduction in oocyst numbers in mosquito midguts. Adsorption onto aluminum hydroxide further enhances immunogenicity. The mechanism of action involves eliciting antibodies against the sexual-stage parasite antigen Pfs25; when mosquitoes ingest blood from vaccinated individuals, these antibodies block parasite development within the mosquito midgut, interrupting malaria transmission[3].
