Pfs25M-EPA + Alhydrogel is a transmission-blocking malaria vaccine candidate composed of a recombinant protein-protein conjugate. The vaccine consists of the Pfs25M antigen—a *Pichia pastoris*-expressed recombinant form of the Plasmodium falciparum surface protein Pfs25—chemically conjugated to ExoProtein A (EPA), a carrier protein derived from *Escherichia coli*. This conjugate is formulated with Alhydrogel, an aluminum hydroxide adjuvant, to enhance immunogenicity. The primary mechanism involves inducing antibodies against the mosquito-stage ookinete surface antigen (Pfs25), thereby blocking parasite development in mosquitoes and interrupting malaria transmission. Developed for intramuscular administration, this subunit vaccine has been evaluated in preclinical and clinical studies for safety and immunogenicity[1][2][4].
