Overview

PfSPZ-GA1 is a genetically attenuated, live malaria vaccine candidate composed of purified, aseptic, cryopreserved Plasmodium falciparum (NF54 strain) sporozoites. These sporozoites are genetically engineered by deletion of the b9 and slarp genes, which arrests parasite development in the early liver stage and prevents progression to blood-stage infection. The vaccine is administered via direct venous inoculation and aims to induce protective immunity against malaria by stimulating both humoral (anti-circumsporozoite protein antibodies) and cellular (interferon-γ–producing T cells) immune responses. Clinical trials have demonstrated that PfSPZ-GA1 is safe and immunogenic in malaria-naïve adults but has shown suboptimal protective efficacy so far.
